Геном высших эукариот содержит огромное количество генов и их регуляторных элементов, таких как энхансеры, сайленсеры и инсуляторы. Энхансеры могут активировать транскрипцию гена независимо от ориентации и, находясь от промотора на расстоянии, которое может достигать нескольких сотен тысяч пар нуклеотидов (Dorsett 1999). Многие энхансеры не имеют специфичности к определенному промотору и. способны активировать транскрипцию с целого ряда генов. Так как в геноме гены располагаются часто достаточно близко друг к другу, то должны существовать механизмы, обеспечивающие взаимодействие энхансера с промотором своего гена и предотвращающие активацию чужих промоторов. В определении взаимодействий между энхансерами и промоторами важную роль играют инсуляторы и сайленсеры. Инсуляторы блокируют взаимодействие между энхансером и промотором, находясь между ними, но, при этом не влияют на их активность, т.е. промотор может быть активирован любым неизолированным энхансером, а энхансер может активировать любой другой неизолированный промотор. В то же время репрессирующее действие сайленсеров не зависит от пространственного расположения по отношению к энхансеру или промотору.

В 1985 году были открыты первые два инсулятора дрозофилы, ses (specialised chromatin structure) и ses', которые окружают ген теплового шока hsp70 дрозофилы и изолируют его от окружающего хроматина (Udvardy et al, 1985). Инсуляторы обнаружены также в регуляторной части гена Abd-B и в мобильном элементе gypsy (инсулятор Su(Hw)). Недавно было показано, что инсуляторы Su(Hw) способны взаимодействовать друг с другом, приводя к выпетливанию участка ДНК, находящегося между ними, и способствуя коммуникации между энхансерами и промотором (Muravyova et al, 2001). В то же время остается неизвестным, все ли инсуляторы способны проявлять данные свойства, а также, способны ли взаимодействовать инсуляторы из разных локусов.

Другой неразрешенной проблемой является выяснение механизма регуляции экспрессии гена Abd-B. Регуляторная область этого гена содержит все три класса регуляторных элементов. Предполагается, что инсуляторы окружают каждый энхансер и организуют ДНК регуляторной области теш Abd-B в серию изолированных хроматиновых доменов.

При этом в непосредственной близости от инсулятора или даже в его составе находятся так называемые PRE-сайты {Polycomb Responsible Elements), участвующие в процессе сайленсинга, необходимого для нормального развития. В результате, регуляторные элементы, находящиеся между хроматиновыми доменами, с одной стороны функционируют как границы, а с другой стороны осуществляют функцию сайленсинга, что позволяет поддерживать индивидуальный и независимый паттерн экспрессии гена Abd-B. Парадоксально то, что энхансер может взаимодействовать с промотором на больших дистанциях, несмотря на присутствие инсуляторов и сайленсеров, и при этом правильно узнавать свой промотор. Таким образом, выяснение механизмов действия инсуляторов и сайленсеров представляет большой интерес, так как позволяет приблизиться к пониманию проблемы взаимодействия между энхансером и промотором в геноме. Кроме того, изучение действия инсуляторов является важной прикладной задачей для создания векторов, которые способны поддерживать стабильную экспрессию генов вне зависимости от места интеграции в геноме.

В данной работе были изучены свойства инсуляторов и сайленсеров из разных локусов, а также их способность регулировать взаимодействия между энхансерами и промотором.

6.ВЫВОДЫ

1. Подтверждены инеуляторные свойства регуляторных элементов ses и zw5 в трансгенной системе генов yellow и miniwhite. Впервые продемонстрирована возможность взаимодействия инсуляторов ses и zw5 друг с другом, приводящая к взаимной нейтрализации их инсуляторной активности.

2. Показано, что ses и zw5 не способны взаимодействовать с инсулятором Su(Hw) и нейтрализовать его активность.

3. Подтверждены инеуляторные свойства регуляторных элементов Fab-7 и Fab-8 в трансгенной системе генов yellow и miniwhite. Показано, что Fab-8, в отличие от Fab-7, способен взаимодействовать с инсулятором Su(Hw) и частично нейтрализовать его активность.

4. Показано, что взаимодействие сайленсеров из регуляторной области гена Abd-B не способно усиливать взаимодействие между инсуляторами.
