Изучение регуляции экспрессии различных генов - важное направление в современной молекулярной генетике. Регуляция экспрессии может осуществляться на нескольких принципиально различных уровнях. К таковым надо отнести уровень регуляции транскрипции, регуляции трансляции, уровень пост-транскрипционных модификаций РНК. Знание о процессах регуляции экспрессии генов позволяет во многом приблизиться к пониманию механизмов функционирования не только клетки, как основной единицы живого, но и многоклеточного организма. Такие процессы как дифференцировка тканей, эмбриогенез, иммунный ответ, образование опухолей и т. д. сводимы на молекулярном уровне к включению одних генов и выключению других. Именно из-за этого изучение регуляции генов является во многом центральным для современной науки. Именно в этой области молекулярная биология с одной стороны, и науки, изучающие такие многоклеточные структуры, как ткани и органы, их системы (гистология, иммунология), организмы, как целое, (физиология, биология развития, эмбриология), с другой стороны, практически смыкаются друг с другом.

Продукт гена tag7 / PGRP -интересен тем, что с одной стороны имея некоторые функциональные черты, близкие такому семейству генов, как цитокины, проявляет с другой стороны пептидогликан-распознающую активность и структурно (по вторичной и прогнозируемой третичной структуре белка) близок к лизоциму. Поэтому, изучение регуляции транскрипции этого гена представляет несомненный интерес как для понимания фундаментальных закономерностей регуляции работы генов, так и для понимания роли tag7/PGRP в функционировании эукариотического организма.

Основной целью данной работы являлось изучение регуляции транскрипции мышиного гена tag7/PGRP на различных модельных системах и выявление и характеристика элементов промоторной области данного гена, ответственных за поддержание транскрипционной активности в клетках тех линий, где наблюдается экспрессия tag7/PGRP и за подавление транскрипционной активности в клетках линий, где экспрессия гена отсутствует. Ставилась также задача изучить возможность индукции данного гена различными агентами в ряде клеточных линий и характеризовать элементы его промоторной области, ответственные за нее.

4. ВЫВОДЫ

1. Впервые определена последовательность промоторной области гена tag7/PGRP с координатами +671, -3534 и проведено картирование участков гиперчуствительности к ДНКазе I, локализующихся в пределах данной области.

2. Показано, что минимальный промотор гена tag7/PGRP, включающий только TATA бокс гена tag7/PGRP с минимальным окружением без вышележащих участков, неактивен во всех исследованных клеточных линиях.

3. На участке с координатами -48, -192, локализован энхансерный элемент, ответственный за положительную регуляцию транскрипции гена tag7/PGRP в клеточных линиях KCMJ10 и BMP-печень. При помощи сайт-направленного мутагенеза впервые показано, что в состав данного энхансерного элемента входят сайты связывания факторов NF-kB и Ets, влияющие на транскрипционную активность гена кооперативно.

4. Показано, что упомянутый выше энхансерный элемент активируется РМА (в клетках линий Jurkat и LBRM) и LPS (в клетках линий WEHI231 и KCMJI0). .На модельной системе клеток линии Jurkat показано, что за индукцию транскрипции гена РМА ответственны сайты связывания факторов NF-kB и Ets.

5. Показано, что мРНК гена tag7/PGRP в клетках KCMJI0 является долгоживущей, время ее полужизни составляет более шести часов и не изменяется под действием LPS.

6. На участке промоторной области с координатами -764, -563, локализован элемент негативной регуляции транскрипции (сайленсер), действие которого зависит от положения, и ответственный за отсутствие транскрипционной активности гена в клеточной линии КСМЛ100. Методом футпринтинга in vitro с использованием ДНК-азы I показано присутствие в составе данного сайленсерного элемента потенциальных сайтов связывания факторов NF-Zz (NF-Zc), NF-1/L и GR.
