Теломеры -это специализированые ДНК-белковые комплексы, находящиеся на концах линейных хромосом, которые гарантируют стабильность эукариотического генома (Pardue and DeBaryshe 1999; Zakian 1996). Существуют специальные механизмы, которые добавляют ДНК к концам хромосом эукариот, балансируя потерю терминальной ДНК, возникающую за счет недорепликация (Blasco et. al. 1999; Pardue and DeBaryshe 1999). У большинства эукариот специальная обратная транскриптаза-теломераза, добавляет теломерные ДНК-повторы к концу хромосомы, используя внутреннюю РНК-матрицу (Blasco et.al. 1999; Pardue and DeBaryshe 1999; Greider 1999). Теломеры Drosophila melanogaster имеют особую структуру. Они состоят из множества копий НеТ-А и TART элементов, принадлежащих к семейству ретротранспозонов не содержащих LTR (Biessmann and Mason 1997; Biessmann et.al. 1997; Mason et.al. 2000; Pardue and DeBaryshe 1999; 2000; Mason et.al. 2000). В телом ерах НеТ-А и TART элементы ориентированы "голова к хвосту" (Biessmann and Mason 1997; Levis et.al. 1993; Walter et.al. 1995).

Несколько лет назад были получены линии Drosophila melanogaster, несущие терминальные делеции (Biessmann and Mason 1988; Biessmann et.al. 1990a; Golubovsky et.al. 2001; Levis 1989; Mason et.al. 1984; Traverse and Pardue 1988). Терминально делетированные хромосомы Drosophila демонстрируют свойства хромосом с нормальными теломерами: они устойчиво поддерживаются в течение многих поколений

Biessmann et.al. 1990а; Levis 1989). Было обнаружено, что НеТ-А и TART элементы способны присоединяться к концам терминально делетированных хромосом (Biessmann et.al. 1990b; 1992; Sheen and Levis 1994; Traverse and Pardue 1988). Частота присоединений

Biessmann et. al. 1992а; Kahn et.al. 2000; Golubovsky et. al. 2001), хотя ничего не известно относительно механизмов, которые контролируют их транспозиции. Также было показано, что терминальные делеции Drosophila могут быть удлинены генной конверсией, использующей в качестве матрицы для конверсии гомологичные теломерные последовательности, или с помощью рекомбинации между теломерными последовательностями (Kahn et.al. 2000; Mikhailovsky et.al. 1999). Однако относительная важность транспозиции мобильных элементов и конверсии в поддержании длины теломер неизвестна.

Основной целью данного исследования было выделение и характеристика нового гена Enhancer of terminal gene conversion (En(tc))„ мутации в котором приводят к удлинению теломер дрозофилы по механизму генной конверсии. Полученные результаты предполагают, что белок En(tc) контролирует удлинение теломер по механизму репликации ДНК. Таким образом, вопреки сложившемуся мнению, теломеры дрозофилы удлиняются не только в результате транспозиции НеТ-А и TART мобильных элементов.

Одновременно терминальные делеции в регуляторной области гена yellow были использованы для изучения взаимодействия между инсуляторами и энхансерами на больших дистанциях. Инсуляторы - это регуляторные последовательности, которые, находясь перед промоторм, способны предотвратить его активацию или репрессию (Corees and Felsenfeld 2000; Bell et.al. 2001). Лучше всего охарактеризованный инсулятор - это 12 сайтов связывания для белка Su(Hw), которые находятся в регуляторной области ретротранспозона МД1 '4.(Mazo et al. 1989; Geyer 1997). Ранее было показано, что Su(Hw) инсулятор полностью блокирует взаимодействие между энхансерами и промотором гена yellow (Geyer and Corees 1992). Например, у мутант (Рис. 14А) индуцирован инсерцией МДГ4 между энхансерами, контролирующими экспрессию yellow в теле и крыльях, и промотором гена (Geyer et. al 1986). В результате энхансеры гена изолированы от промотора и их взаимодействие блокировано инсулятором. В Gause et al. (1998) была

2 rhl описана производная у мутации у , которая содержала дупликацию регуляторной области гена yellow и МДГ4. Было показано, что дупликация МДГ4 элемента полностью подавляет инсуляцию yellow промотора, отделенного от энхансеров тела и крыльев

Su(Hw) инсулятором (Рис.14Б). Целью данного исследования было выяснение причины подавления инсуляции. Для этого была получена коллекция yellow аллелей, имеющих концы терминальных делеций в разных частях регуляторной области угМ аллеля. В результате было обнаружено, что Su(Hw) может блокировать взаимодействие между yellow энхансерами и промотором, если терминально делетированная хромосома заканчивается дистальнее энханссров yellow . Кроме того, по крайней мере, 5 из 12 сайтов связывания для Su(Hw), содержащихся в Su(Hw) инсуляторе, расположенном за энхансерами yellow, позволяют преодолеть блокирующее энхансеры действие Su(Hw) инсулятора, встроившегося между энхансерами и промотором гена yellow. На основе полученных результатов была предложена модель, по которой два или несколько соседних инсуляторов взаимодействуют друг с другом, блокируя или облегчая энхансер-промоторные взаимодействия, в зависимости от конфигурации других ¿/г/с-регуляторных элементов (энхансеров, сайленсеров и промоторов).

5. ВЫВОДЫ.

1. Идентифицирована и картирована новая мутация, En(tc), которая увеличивает частоту терминальной генной конверсии.

2. Показано, что теломеры дрозофилы удлиняются не только в результате транспозиций мобильных элементов, но и по механизму терминальной генной конверсии.

3. Продемонстрировано, что Su(Hw) инсулятор может блокировать энхансеры даже если находится на конце хромосомы, несущей терминальную делецию.

4. Доказано, что два Su(Hw) инсулятора не формируют изолированный транскрипционный домен. Энхансеры гена yellow, окруженные Su(Hw) инсуляторами, могут эффективно активировать изолированный промотор.
